He trained under the Warrior's scholarship scheme. Gleeson played in the U18s and U21s grand finals for Wigan in 2005. He scored for the U18s and helped them win against Leeds Rhinos.

He made his debut for Wigan's first team in 2005 and played three games in that season. He was awarded man of the match in one game. He also played for the Warriors in the match against Harlequins in 2006. During his time at Wigan Warriors, Sean was loaned to Widnes Vikings in order to give him more experience of being in the first team.

Sean is just one of three Gleeson family members who play rugby league. His cousins Mark and Martin both play for Warrington Wolves.

He can play in either centre position or at full back.
